CristalO - crystallography core facility

Our platform’s core mission is to support, monitor, and oversee projects that rely on protein-applied crystallography. All our services are offered within the EMBRC-France infrastructure.

Crystallogenesis and Structure Determination

The crystallogenesis service involves producing crystals from a soluble protein. Crystallization conditions are automatically identified using a crystallization robot before the manual steps of reproducing and optimizing the crystals.

Structure determination is then performed using protein crystallography, which consists of exposing the crystals to synchrotron X-rays to obtain diffraction, collecting the data, and carrying out the resolution and refinement of the 3D coordinates.

  • Determination of the oligomeric state of a sample by DLS
  • Sample crystallization using a Crystal Gryphon crystallization robot
  • Monitoring of crystallization with a CrysCam camera and/or a stereomicroscope
  • Manual reproduction and optimization of crystallization conditions
  • Determination of cryo-freezing conditions for crystals
  • X-ray diffraction of crystals
  • Data collection and processing
  • Phasing, reconstruction, and refinement of the 3D structure

Le robot nano-goutte Gryphon automatise la préparation de gouttes de cristallisation à très faible volume dans des plaques 96 puits. Il utilise la technique de diffusion en phase vapeur en goutte assise, garantissant précision, rapidité et reproductibilité dans le criblage des conditions de cristallisation.
